{"id":47556,"date":"2014-08-25T22:39:47","date_gmt":"2014-08-25T20:39:47","guid":{"rendered":"http:\/\/staging.sillon38.com\/?p=47556"},"modified":"2014-08-25T22:39:47","modified_gmt":"2014-08-25T20:39:47","slug":"le-genome-du-colza-sequence","status":"publish","type":"post","link":"https:\/\/www.sillon38.com\/blog\/2014\/08\/25\/le-genome-du-colza-sequence\/","title":{"rendered":"Le g\u00e9nome du colza s\u00e9quenc\u00e9"},"content":{"rendered":"<div>\n<div>\n<div><img decoding=\"async\" title=\"Inflorescence de colza. \u00a9 CAIN Anne-H\u00e9l\u00e8ne \" src=\"https:\/\/inra-dam-front-resources-cdn.brainsonic.com\/ressources\/afile\/169323-2d1ce-picture_client_format_0-8031-0001-jpg.html\" alt=\"Inflorescence de colza. \u00a9 CAIN Anne-H\u00e9l\u00e8ne \" \/><\/div>\n<\/div>\n<div>\n<p><span style=\"color: #000080;\">Un consortium international d\u2019une trentaine d\u2019instituts de recherche,  pilot\u00e9 par l\u2019Inra et le CEA (Genoscope) et associant le CNRS et  l\u2019universit\u00e9 d\u2019Evry, vient de rendre publique la s\u00e9quence de r\u00e9f\u00e9rence  du g\u00e9nome complet du colza. <\/span><\/p>\n<p><span style=\"color: #000080;\">Cette premi\u00e8re mondiale ouvre la voie \u00e0 la  compr\u00e9hension fondamentale des g\u00e9nomes complexes polyplo\u00efdes (associant  plusieurs sous-g\u00e9nomes), ainsi qu\u2019\u00e0 l\u2019am\u00e9lioration vari\u00e9tale du colza,  une grande culture r\u00e9cente<sup>1<\/sup> en pleine expansion et \u00e0 fort potentiel d\u2019am\u00e9lioration. <\/span><\/p>\n<h2><span style=\"color: #000080;\">Une premi\u00e8re mondiale<\/span><\/h2>\n<p><span style=\"color: #000080;\">Dans le cadre du projet Seq-Poly-Nap,  financ\u00e9 principalement par l&rsquo;Agence nationale de la recherche (ANR), des  chercheurs de l\u2019Inra, du CEA (Genoscope), du CNRS, de l\u2019Universit\u00e9  d\u2019Evry, en collaboration avec leurs coll\u00e8gues \u00e9trangers, ont produit la  s\u00e9quence de r\u00e9f\u00e9rence du g\u00e9nome du colza ainsi que celle d\u2019une  collection de vari\u00e9t\u00e9s repr\u00e9sentant la diversit\u00e9 de cette esp\u00e8ce. <\/span><\/p>\n<p><span style=\"color: #000080;\">Une  s\u00e9quence de r\u00e9f\u00e9rence constitue la s\u00e9quence assembl\u00e9e et ordonn\u00e9e de  tous les g\u00e8nes. La principale difficult\u00e9 dans le cas du colza a \u00e9t\u00e9 de  s\u00e9quencer les deux sous-g\u00e9nomes et de les distinguer. Ceci a \u00e9t\u00e9 rendu  possible par la mise au point d\u2019une strat\u00e9gie originale de s\u00e9quen\u00e7age,  le d\u00e9veloppement d\u2019outils bioinformatiques et d\u2019analyse de l\u2019expression  des g\u00e8nes dupliqu\u00e9s et de leur r\u00e9gulation.<\/span><\/p>\n<p><span style=\"color: #000080;\">C\u2019est la premi\u00e8re fois  qu\u2019un g\u00e9nome polyplo\u00efde r\u00e9cent est s\u00e9quenc\u00e9 dans son int\u00e9gralit\u00e9 et  compar\u00e9 \u00e0 ceux de ses esp\u00e8ces parentales, le chou et la navette (dont  l\u2019Inra a \u00e9galement contribu\u00e9 aux d\u00e9chiffrages r\u00e9cents).<\/span><\/p>\n<p><span style=\"color: #000080;\"><a id=\"eztoc3356237_2\" name=\"eztoc3356237_2\"><\/a><\/span><\/p>\n<h2><span style=\"color: #000080;\">Le colza: champion des duplications r\u00e9currentes des g\u00e9nomes par polyplo\u00efdie<\/span><\/h2>\n<p><span style=\"color: #000080;\">Les  chercheurs montrent qu\u2019au-del\u00e0 de l\u2019hybridation post-n\u00e9olithique ayant  conduit \u00e0 sa formation, le colza aurait accumul\u00e9 au cours de son  \u00e9volution 72 g\u00e9nomes ancestraux, r\u00e9sultat de nombreux cycles de  polyplo\u00efdisation, faisant de son g\u00e9nome un des plus hautement dupliqu\u00e9s  chez les plantes \u00e0 fleurs (angiospermes). <\/span><\/p>\n<p><span style=\"color: #000080;\">Ce ph\u00e9nom\u00e8ne r\u00e9current, suivi  par des restructurations du g\u00e9nome, a conduit \u00e0 l\u2019accumulation d\u2019un  grand nombre de g\u00e8nes, soit plus de 101 000. Un nombre plus de quatre  fois plus important que les 20 000-25 000 g\u00e8nes de l\u2019Homme par exemple.<\/span><\/p>\n<p><span style=\"color: #000080;\"><a id=\"eztoc3356237_3\" name=\"eztoc3356237_3\"><\/a><\/span><\/p>\n<h2><span style=\"color: #000080;\">Une cohabitation et un \u00ab dialogue \u00bb subtils entre les sous-g\u00e9nomes et g\u00e8nes dupliqu\u00e9s<\/span><\/h2>\n<p><span style=\"color: #000080;\">Les  chercheurs ont observ\u00e9 que la grande majorit\u00e9 des g\u00e8nes du colza sont  dupliqu\u00e9s, existant donc en deux copies \u00e0 s\u00e9quences proches ou  quasi-identiques. La quasi-totalit\u00e9 de ces g\u00e8nes dupliqu\u00e9s sont exprim\u00e9s  ; pour un m\u00eame g\u00e8ne, les deux copies participent conjointement \u00e0 leur  fonction. <\/span><\/p>\n<p><span style=\"color: #000080;\">Les chercheurs sugg\u00e8rent que ces g\u00e8nes dupliqu\u00e9s conf\u00e8rent un  r\u00e9servoir important de diversification, d\u2019adaptation et d\u2019am\u00e9lioration;  la fonction principale \u00e9tant r\u00e9gie par une copie des g\u00e8nes dupliqu\u00e9s, la  deuxi\u00e8me copie peut se restructurer et muter pour l\u2019\u00e9mergence de  nouvelle fonction.<\/span><\/p>\n<p><span style=\"color: #000080;\">Le dialogue se traduit \u00e9galement par des  \u00e9changes de g\u00e8nes et d\u2019ADN entre les deux sous-g\u00e9nomes du colza. Ainsi,  pour un g\u00e8ne dupliqu\u00e9, normalement pr\u00e9sent sur les deux sous-g\u00e9nomes,  une copie peut \u00eatre remplac\u00e9e par la s\u00e9quence de la deuxi\u00e8me copie du  second sous-g\u00e9nome. Le m\u00e9canisme d\u2019\u00e9change \u00e0 l\u2019\u0153uvre ainsi que son  avantage s\u00e9lectif restent encore \u00e0 d\u00e9terminer, bien qu\u2019il ait d\u00e9j\u00e0 \u00e9t\u00e9  montr\u00e9 que ce ph\u00e9nom\u00e8ne conduit \u00e0 une diversification.<\/span><\/p>\n<p><span style=\"color: #000080;\"><a id=\"eztoc3356237_4\" name=\"eztoc3356237_4\"><\/a><\/span><\/p>\n<h2><span style=\"color: #000080;\">Une ressource unique pour l\u2019am\u00e9lioration vari\u00e9tale du colza<\/span><\/h2>\n<p><span style=\"color: #000080;\">Le  colza est une esp\u00e8ce cultiv\u00e9e \u00e0 grande \u00e9chelle depuis peu, \u00e0 laquelle  il reste un fort potentiel d\u2019am\u00e9lioration g\u00e9n\u00e9tique. Ainsi le s\u00e9quen\u00e7age  de son g\u00e9nome constitue une ressource unique au monde car il ouvre de  nouvelles potentialit\u00e9s pour l\u2019identification des g\u00e8nes d\u2019int\u00e9r\u00eat  agronomique et leur utilisation rapide dans les programmes de s\u00e9lection  vari\u00e9tale. <\/span><\/p>\n<p><span style=\"color: #000080;\">Il serait par exemple possible d\u2019am\u00e9liorer la teneur et la  composition en huile, la r\u00e9sistance \u00e0 des pathog\u00e8nes, la tol\u00e9rance au  froid, le rendement, ou encore l\u2019efficacit\u00e9 d\u2019utilisation des nitrates  dans le sol.<\/span><\/p>\n<p><span style=\"color: #000080;\">De nombreux projets exploitant cette ressource pour une agriculture durable sont en cours, notamment \u00e0 l\u2019Inra.<\/span><\/p>\n<p><span style=\"color: #000080;\"><sup>1<\/sup> L\u2019apparition  du colza est consid\u00e9r\u00e9e comme \u00ab r\u00e9cente \u00bb, c\u2019est-\u00e0-dire qu\u2019elle remonte  \u00e0 l\u2019\u00e8re post-n\u00e9olithique, soit environ 5 500 ans avant notre \u00e8re<\/span><\/p>\n<p><span style=\"color: #000080;\"><span style=\"color: #ff6600;\"><strong>R\u00e9f\u00e9rence<\/strong><\/span><br \/>\nChalhoub <em>et al<\/em>. <strong>Early allopolyploid evolution in the post-Neolithic <em>Brassica napus<\/em> oilseed genome<\/strong>. <em>Science<\/em>, 22 ao\u00fbt 2014.
